Mechanism of Action :
Inhibits protein synthesis in bacterial cell by binding reversibly to the P site on the 50S subunit of the bacterial ribosome. This action is considered to be bacteriostatic

INDICATIONS:
Gonorrhea caused by penicillin-resistant bacteria or in patients hypersensitive to penicillins.
DOSAGE AND ADMINISTRATION
Adults:
Deep I.M injection as a single dose (Spectinomycin 2g)
A dose of 4g may be required divided between two injection sites
Mutiple-dose courses have been used for disseminated Infections
Children over 2 years :
Spectinomycin 40mg/kg stat.
PRECAUTIONS
Nausea, Dizziness, fever, chills , insomnia and urticaria have ocassionally occured with single doses of Spectinomycin.
Anaphylaxis has occured rarely.
Mild to moderate pain has been reported after I.M injections.
Alterations in Kidney and liver functions and a decrease in haemoglobin and haematocrit values have occassionally been seen with repeated doses
Although a reduction in urine output has been seen after single and multiple doses , Spectinomycin has not been noted to produce functional indicative nephrotoxicity.
CONTRAINDICATIONS:
Hypersensitivity to Spectinomycin or Macrolides.
DRUG INTERACTIONS:
Lithium toxicity has been reported on isolated occassions in patients receiving spectinomycin.
SIDE EFFECTS
Urticaria
Dizziness
Nausea
Chills
Fever
Decrease in Hemoglobin.
