Nifuroxazide Mode of Action:
Nifuroxazide inhibits the constitutive phosphorylation of STAT3 in MM cells by reducing Jak kinase autophosphorylation, and leads to down-regulation of the STAT3 target gene Mcl-1.
INDICATIONS
Nifuroxazide belongs to the group of the intestinal anti-infective drugs. It is available as an over the counter medication for the treatment of acute diarrhea if it is caused by bacteria. Studies have shown that this medication reduces the number of stools after only two days of treatment and reduces the duration of diarrhea for around twenty four hours. Nifuroxazide does not affect the intestinal flora
DOSAGE AND ADMINISTRATION
The usual dose for children aged 6-18 years is 200 mg three times a day. In adult patients, the recommended dose is 200 mg four times a day. Duration of therapy should not be longer than seven days.
In children aged 2-6 years Nifuroxazide should be taken as oral suspension 200 mg / 5 ml. Recommended dose is 5 ml three times a day. Use of this drug in children younger than 2 years is not recommended.
This drug can be taken regardless of meals.
CONTRAINDICATIONS / PRECAUTIONS:
Contraindication to the use of the Nifuroxazide is hypersensitivity to this substance.
Nifuroxazide should not be taken in the following situations:
- If you have bloody stools
- If you have a fever
- If you are vomiting
DRUG INTERACTIONS:
Nifuroxazide should not be administered concomitantly with following drugs:
- Metronidazole (a drug used in the treatment of anaerobic infections and infections caused by parasites)
- Sulfonamides (e.g. Bactrim), which is mainly used to treat urinary tract infections
- Cephalosporins (antibiotics)
- Chloramphenicol
- Nitrofurantoin (a drug used to treat urinary tract infections)
- Griseofulvin – a drug used in the treatment of fungal infections
Nifuroxazide should never be administered in combination with the above mentioned drugs as this increases the risk of disulfiram-like reaction, which is characterized by: severe nausea and vomiting, headache and rapid heartbeat.
Also, this drug should not be administered simultaneously with sedatives (e.g. benzodiazepines, such as: bromazepam, alprazolam, lorazepam, midazolam and others).
ADVERSE DRUG REACTIONS:
Nifuroxazide may cause the following side effects:
- Quincke’s edema (angioneurotic edema which includes subcutaneous tissue and is characterized by allergic reaction)
- Urticaria
- Anaphylactic shock
